Another fantastic "what car?" post.
What would you do (as opposed to what would scooby do)
£20k to £25k
New or used
2 doors (4 if it was something like an M5 )
Rear seats - nice to have but not important
Must do 0-60 in less than 6.5
Must NOT be a vauxhall
Insurance group irrelevant
Fuel economy >25MPG on a long motorway run (ave 80/90 MPH)
full of bells and whistles i.e aircon leather (cruise control)
I have a list in mind of such like as used M5 / M3 Merc SL500 / BM 330 coupe or conv etc etc
anything else I should look at ??

If it was me, I'd buy two cars
One comfy barge and one weekend\trackday toy. That way, the 'toy' car can be as impractical\modified as you like, and you can have the comfy car for going to-and-from work.
This isn't practical for everyone 'cos you need to house two cars, but I do it for many reasons, such as if you have one valuable car, you have to pay it's running expenses inc. depreciation as the miles pile on. You're subject to Tesco's parking dents, scratches etc.
Just an idea for ya
I currently have a 3.0 X-Type SE and an MR2 Turbo T-Bar approaching 400bhp Although TBH, I'd rather change the Jag for something cheaper and spend more on the MR2

I've been running 2/3 cars for a while now - just want something to rip up and down the motorway and yet be fun to drive.. until I decide on a nice 993/911

Would have thought that you would have been very hard pushed to get >25mpg out of a C43 AMG or SL500.......bloody nice cars though, how about a C32AMG? might stand a better chance with that, although im not sure if they would fall in to your bracket, cant argue with 345 horses of pure stealth though

OOF - nawp to the Lexus as they are a bit bland to drive etc..
Neilo the C43 AMG will do 25 (just) on a long run.. but if you hoon it around town you will get single figures

My C43 did 27 mpg the other week on a long run - more like 17 on short commute. Great car though - lovely V8 noise when pushed. C32 is the new shape equivalent but struggle to get them below £27-28k.

A last of the line toyota supra twin turbo with vvt and a 6sp box, I think the imports were made till late 99 or 2000??? very reliable very smooth and fecking brutally quick!!

Mondeo ST220....
I know of a black 04 plate saloon with about 1k miles and EVERY extra on the option list added to it (leather, sat nav, air con + ERS, privacy glass). Mail me if you want more details.... Cost ~£26k about a month ago....
Alot more car for the money compared to say, a BMW 330i

Yeah, the mondeo is popular... but the ST220 looks sleeker, and not too many of them on the go. Well, not up here anyway! I drove this one and loved it. Felt so solid & powerful on the road, and that was driving it on some twisty roads in the lake district
It is just out of my price bracket

Drove an RX8 lo-power last night. MIGHTY impressed. So much so that I have spent all day trying not to pick up the phone and get a hi-power on order. Seems to fit your requirements nicely, although MPG could be a problem.
I've currently got one on your list, an Mercedes SL500. Fantastic car, but struggling to get more than 16MPG.
My other "must own one day" cars that could fit are:
Porsche 968 Club Sport
Porsche 911 Carerra 2 (manual)
Merc E55AMG
Beemer M Coupe or M5
