Try a different coil pack, Take one you know works and swap them over if the problem follows that coil pack theres your problem might be easier to swap the front two over, They are easy to unplug just follow the lead till you find the multi plug unplug and swap, If the problem stays on that piston and you say you put a new plug in it might be the other end of the lead.

But to be honest like Joz said it does sound like THAT coil pack has come to the end of its life, maybe the end has broken, its common for the plastic ends to brake.

The engine ecu would supply the coil. I would of thought it should have a feed and a switched earth as the driver for the coil. So check the wiring to the coil>

I would also be inclined to check the compression on that cylinder.

Did you try swapping the coilpacks as jaywalker suggested?
If you have a good spark at the plug I would check fuel supply. Is the plug wet? If not maybe the injector connection/wire is faulty or fuel rail blocked.
