By far and away the most comprehensive anti-spyware software just now is the Microsoft malicious software removal tool.

If any nasty software is running on your PC, it'doesn't just delete what files it thinks are infected (like adaware, spybot etc) it comrehensively removes all associated traces to prevent them from re-surfacing.
