Can't open programmes in XP Pro

Default Can't open programmes in XP Pro

Since WIndows update installed a service pack, I think, into XP Pro, a couple of programmes won't run. Whenever I try to open them I get the following with 'Close' & 'Ignore' buttons. Neither button does anything.

"16 bit Windows Subsystem"
"C:\WINDOWS\SYSTEM32\AUTOEXEC.NT. The system file is not suitable for running MS-DOS and Microsoft Windows applications. Choose 'Close' to terminate the application"

Any suggestions for getting round this problem? One prog. is an old cardfile which it would be nice to get back again.
