Try temporarily disabling your firewall and anti-virus. If this works, open the SMTP mail ports in your firewall.

Does your ISP require secure logging ?

Ive had this problem when using Sygate Personal Firewall, never did fix it satisfactorily but suspect that manually opening the ports was the solution.

There is an option in Outlook thats something along the lines of "require SSL" which when checked will only send using SSL encryption. If your ISP requires this then that could be why your e-mails arent sending. Which ISP are you with btw?
