Via the M3, from my experience it usually takes around 45 minutes to get to Winchester from the M25, assuming an average amount of traffic. After 8pm though, you could be looking at perhaps 35 minutes, as it's quieter then. Look out for various roadworks on the M3 though and 50mph limits (yes, there is a speed camera or two, so watch out), so I'd allow an hour or so from the M25 (jct 12), plus however long it'll take you to get to that point at your end. Then maybe 5-10 minutes to get into town (Winchester) from the M3.
If you want to avoid the M3, you could take the A31 from Guildford, which is a more interesting route but will take you a bit longer - about 45-60 minutes from Guildford depending on traffic.
When I used to drive home from the London area, during the normal evening rush hour, the worst traffic was always on (and north of) the M25 - once south of the M25, I always found it was better. Heavy traffic, but all moving at a reasonable pace.
